چکیده

Background: Pathological (or heterotopic) calcification is the deposition of calcium-containing crystals in soft tissues that normally do not calcify. The these tendons such as rotator cuff and Achilles tendon known calcific tendinopathy (CT). CT a painful condition, which increases rupture rate leads to disability. Objectives: To understand what inhibits calcification, order provide new strategies treat condition for existing therapies are ineffective. Methods: We investigated role gasotransmitter hydrogen sulfide (H 2 S), particular H S-producing enzyme cystathionine γ-lyase (CSE) CT. In vitro , we induced tenocytes from WT CSE KO mice or treated with different S donors. vivo, was assessed surgery-induced murine model (tenotomy tendon) spontaneous (aging). Samples obtained patients were also analyzed. investigate underlying mechanisms CSE-H effect, focused on bone morphogenic proteins (BMPs) pathway. additionally explored if altered extracellular matrix (ECM) organization, due lysyl oxidase (LOX) activity aberrant collagen-crosslinks, could be involved this context, studied affect LOX expression activity. Results: tenocyte inhibited by exogenous S-donors, while it exacerbated tenocytes. protective confirmed vivo. aged mice, microtomography analysis revealed compared WT. CT, an inverse correlation between operated seen over time. Similarly, inversed found human samples. Reduced exposed accompanied decreased genes coding BMP2, BMP4 activation BMP signaling pathway (pSMAD1/5/8). This pointed potential beneficial inhibition, therefore By LOXs gene calcifying much higher (4-fold) LOX, LOL2 LOXL4 Moreover, S-donors Altogether, results suggest lead activity, excessive collagen cross-links ECM, ultimately Further experiments ongoing prove hypotheses. Conclusion: targeting production CSE, supplying S-donor, therapeutic relevance pathological context can modify its disease course. anti-mineralizing effect both inhibition suppression abnormal Disclosure Interests: None declared.

Epithelial Electrolyte Transport Physiology and the Gasotransmitter Hydrogen Sulfide

Hydrogen sulfide (H2S) is a well-known environmental chemical threat with an unpleasant smell of rotten eggs. Aside from the established toxic effects of high-dose H2S, research over the past decade revealed that cells endogenously produce small amounts of H2S with physiological functions. H2S has therefore been classified as a "gasotransmitter." A major challenge for cells and tissues is the m...

Blood pressure control: hydrogen sulfide, a new gasotransmitter, takes stage.

Hydrogen sulfide (H2S) is the most recently characterized autocrine/paracrine messenger implicated in the control of vascular tone. A series of coherent observations now document that this gas is a strong vasorelaxant and a determinant of blood pressure in experimental models. Targeted deletion of the gene encoding cystathionine-lyase (CSE). CSE, a key enzyme for H2S biosynthesis, reduces serum...

Protective Mechanism of Hydrogen Sulfide against Chemotherapy-Induced Cardiotoxicity

Over the past few decades, the number of long term survivors of childhood cancers has been increased exponentially. However, among these survivors, treatment-related toxicity, especially cardiotoxicity, is becoming the essential cause of morbidity and mortality. Thus, preventing the treatment-related adverse effects is important to increase the event free survival during the treatment of cancer...

Hydrogen sulfide (H2S) - the third gas of interest for pharmacologists.

Nitric oxide (NO) and carbon monoxide (CO) synthesized from L-arginine by NO synthase and from heme by heme oxygenase, respectively, are the well-known neurotransmitters and are also involved in the regulation of vascular tone. Recent studies suggest that hydrogen sulfide (H(2)S) is the third gaseous mediator in mammals. H(2)S is synthesized from L-cysteine by either cystathionine beta-synthase...

Hydrogen sulfide (H2S) releasing agents: chemistry and biological applications.

Hydrogen sulfide (H2S) is a newly recognized signaling molecule with very potent cytoprotective actions. The fields of H2S physiology and pharmacology have been rapidly growing in recent years, but a number of fundamental issues must be addressed to advance our understanding of the biology and clinical potential of H2S in the future. Hydrogen sulfide releasing agents (also known as H2S donors) ...
